The film begins with Columbus (played by Jesse Eisenberg), a neurotic college student, narrating the rules for surviving in a world overrun by zombies. He meets Tallahassee (played by Woody Harrelson), a tough and resourceful zombie killer who is on a quest to find the last Twinkie on Earth.
